For example, put 1.5 mL tubes in a container with a piece of autoclave tape on top of the lid. Do not seal the lid of the container tight -- leave it loose. If the container you are using for autoclaving does not have a lid, use aluminum foil as a lid, and place a piece autoclave tape on top of the foil. For autoclaving pipette tips in a pipette tip box, the lid can be closed; again add a piece of autoclave tape to the lid. If the autoclave tape does not change color after autoclaving (we purchased a defective batch), add a line with a sharpie on the tape after autoclaving and once cooled.
Add your name and information to the sign-up sheet in the autoclave room. Set the autoclave to 121 C. Also set: sterilization time for 15 min and dry time for 10 min. Turn the autoclave off when done and wait until the psi reaches 0 before opening the door (this could take up to 30 min.).
