During the first round of trichome measurements, we noticed that newer leaves had more trichomes than the ones we had measured. Thus, we conducted a second round of trichome measurements (and leaf size) to better characterize trichome density across the plants. This was done on May 29th (reps 1-4) and 30th (reps 5-7), 2018. For each plant, we first found the longest branch (presumably the primary branch). Then, starting from the top, we went down the branch until we found the first undamaged, fully formed leaf that was large enough to measure trichomes (big enough for the stencil to not cross the midvein). We then took measurements as previously described (trichomes 1).
A few notes:
Abbie couldn't find her stencil, so she used mine (ZG). She put her initials as ZG as well so that the proper stencil area could be used. ZG did not actually make any measurements, so this properly accounts for measurer biases as well.
Shay said that a few times she simply couldn't find a big enough leaf. She then let the stencil cross the midvein but didn't count the trichomes on the midvein. Not sure what to do about this yet. We could maybe subtract on the midvein area, or just not use these (she made a note).
We also recorded the distance from the top bud (as with plant height) to the leaf used in case we want to control for this.
We will probably want chemistry, dry weight and SLA on these leaves too. They have been dried and all envelopes have a circled 2 on top.
