M. truncatula plant structure seems to be the same as M. sativa, except in M. sativa, a crown produces multiple stems (see . So, we will measure these traits as we did last year:
We took the second true leaf from the plant. It has three leaflets. We took length and width at widest point (in mm) with calipers of the middle leaflet. We then placed the middle leaflet under a stereoscope, put a circle stencil to the right of the midvein (in the middle of that half of the leaflet) and counted the number of trichomes within the circle. I took the area of each of the seven stencils we used in imageJ (in square pixels) so that we can use them to calculate density.

*Skylar noticed early 17 May that the newer leaves had more trichomes. He counted 30 within the circle on one of them. So, we might repeat this protocol with a newer leaf next week.
