Wed and Thurs of this week (May 31 for reps 1 and 2, June 1 for reps 3-5; this means 20 days after planting), we phenotyped the plants. We took the second true leaf from the plant ("1") off the main branch (B0); see attached leaves.jpeg. It has three leaflets. We took length and width at widest point (in mm) with calipers of the middle leaflet. We then placed the middle leaflet under a stereoscope, put a circle stencil to the right of the midvein (in the middle of that half of the leaflet) and counted the number of trichomes within the circle. I took the area of each of the seven stencils we used in imageJ (in square pixels) so that we can use them to calculate density.
We then saved the entire leaf (three leaflets) in a labeled coin envelope in a bin with desiccant for dry leaf weight (on the middle leaflet) and IR chemistry later.
For this we bought:
coin envelopes
bins for holding dessicant and coin envelopes
dessicant (going to look at Michaels)
6 stencils that have a small hole for trichome density measurement (smaller than a hole punch)
