Making Agar

Agar Timeline:

• Ask the other teachers if they will be needed agar too.

• 3 days to make plates.

• Put Agar plates on the heater to warm then up (Winter).

• Agar nutriment to left in cupboard under the microwave,top left.

• Conical flask to make up agar (Recipe book under Nutriment Agar 7g/250ml tap water).

• 1 plate has 15ml (24 plates has 360ml(15*24=360) ).

• Wipe the bench with pink Virkon (Should be very pink).

• Get a water bath out, fill a quarter with tap water, set to 45 degrees.

• Make the Agar, add the powder slowly making sure everything is dissolved. (can use warm water).

• Place the folded cotton wool in the neck of the cylinder, and cover this loosely with tin foil.

• Autoclave, (see autoclave)

• Place flasks in the water bath after sterilising. Should use a Water bath set at 50oC to let the ager reset for 20/30 mn before pouring

• Lay the warmed plates out, pour the Agar to cover the base of the dish, swirl it around and put back in towers to cool slowly(less condensation)

• To fill specimen bottles(mckenzie bottle) with agar, fill at an angle to a third of the 10ml bottle.

Seeded Plates:

• Same as above, apart from add the bacteria and the broth, instruction on broth bottle in fridge,leave out for a couple of days and cover loosely

• Add a ml of bacteria directly into the 45 degrees Agar in the conical flask while in the water bath and then add the broth.Pour the dish as normal. c

• Put out AB disc,tweezers, sellotape, No.of dishes as agar, loops each, half a dozen specimen bottle, splints, lighter and scissors.
