Functions of Common Electrophysiology Solutions
I+I: isoproterenol and IBMX
Isoproterenol
B2AR agonist
Increases cAMP
IBMX
Phosphodiesterase (PDE) inhibitor
PDE normally degrades cAMP and cGMP
Increases cAMP and cGMP
Role of cAMP
Activates protein kinase A (PKA)
PKA can phosphorylate CFTR
Phosphorylation of CFTR activates/opens it
Activates other ion channels as well
Overall: used to activate ion channels
2ME: 2-mercaptoethanol
Reduces disulfide bonds
Helps stabilize and maintain activity of protein
Reduces variability of results by making the CFTR proteins homogenous
DTT:
Reduces disulfide bonds (more effectively than 2ME)
This is so we have a homogeneous population of the channels in the oocyte, so we can generalize results to a single channel and reduce variability in the results
GlyH101
CFTR inhibitor (dose dependent)
Open channel blocker
SCN-
Blocks CFTR
PEA
Trace amine
Activates TAAR1
MTS reagents
Good experiment
Nothing happens in FR (stable, horizontal line)
I+I: channels activated, conductance increases (no more than ~100ish microsiemens), line goes up, plateau
