Transformation was done on 5/27/21 on LB+AMP plates made on 5/27/21 by AH. Transformation done by HS.
KCNJ3-2 pGHE with a concentration of 10 ng. DNA from Lane 2 on LMC052421B.
This plate shows that the DNA is circular and transformation occurred.
KCNJ3-2 pGHE with a concentration of 1 ng. DNA from Lane 2 on LMC052421B.
This plate shows that the DNA was circular and that transformation occurred. We are unsure as to why the 1ng plate has more colonies than the 10ng plate, but we intend to investigate the cause.
KCNJ3-1 pGHE with a concentration of 10 ng. DNA from Lane 3 on LMC052421A.
This plate shows that the DNA is circular and transformation has occurred.
KCNJ3-1 pGHE with a concentration of 1 ng. DNA from Lane 3 on LMC052421A.
This plate shows that the DNA was circular and that transformation occurred. We are unsure as to why the 1ng plate has more colonies than the 10ng plate, but we intend to investigate the cause.
KNCJ3-2 (assuming pGHE) with a concentration of 10 ng. DNA from Lane 9 on LMC052421A.
This plate shows that the DNA is circular and transformation has occurred.
KCNJ3-2 (assuming pGHE) with a concentration of 1 ng. DNA from Lane 9 on LMC052421A.
This plate shows that the DNA is circular and transformation has occurred. The colonies for both plates are relatively similar, but it does seem that this plate has less colonies than the 10ng, which makes sense as 1ng is a lower concentration.
KCNJ9 pGHE-2 with a concentration of 10 ng. DNA from Lane 10 on LMC052421A.
This plate shows that the DNA is circular and transformation has occurred. There was a possibility before incubation that the KCNJ9 DNA was going to be too diluted for this plate, but it seems that was not a problem based on the colonies seen on this plate.
KCNJ9 pGHE-2 with a concentration of 1 ng. DNA from Lane 10 on LMC052421A.
This plate shows that the DNA is circular and transformation has occurred.
OPRM1-1 pGHE with a concentration of 10 ng. DNA from Box 18.
This plate shows that the DNA is circular and transformation has occurred. We believe the reason that this plate has less colonies is due to too little DNA making it into the tube containing the bacteria before incubation. This was due to the use of the wrong pipette tip to transfer the DNA.
OPRM1-1 pGHE with a concentration of 1 ng. DNA from Box 18.
This plate shows that the DNA is circular and transformation has occurred.
Negative control was MBG water.
This plate is the negative control which contained MBG water. This shows that the transformation was successful as no bacterial colonies grew on this plate.
