Clean bio safety hood with ethanol
Switch on blower, and sterilizing switch should be at fluorescent
Bring supplies to hood
Bottle of LB Broth + Amp (100 ng/µL in Fridge A)
Four Erlenmeyer flasks labeled with KCNJ5_FLAG_pcDNA3.1 [Concentration] MN and AJH
Clean gloves with ethanol and allow to dry
Pour 75 mL of broth into each flask and 75 µL of AMP
Use a pipette tip to remove an acceptable (predetermined) colony GENTLY as not to scar the agar
Medium sized and isolated from other colonies
Drop the pipette tip STRAIGHT down into its corresponding flask
Return the lid to the plate and the foil to the flask and set aside
Repeat for all flasks
After all flasks have been inoculated, place in the small incubator (SHAKIRA SHAKIRAAA) at 37ºC and shake at 170 rpm for 15-16 hours.
Replace materials and clean work area; shut off the hood light and blower
COMPLETE AT 11:16PM

Broth is not cloudy, indicating there was no contamination of the broth, micropipette tips, or Erlenmeyer flask.
