Freezing Protocols
Vitrification
Slow freezing
4/19/18
Vitrification solutions made with the following amounts:
10 mL total solution with 5M DMSO and 1M Mannitol
DMSO - 3.55 mL
Mannitol - 1.8035 g
MBSH Water - 6.45 mL
With this 10 mL solution, 6 1.5 mL tubes were loaded with 1 mL each. 3 of these tubes placed in freezer C, box 5 (labeled “CV”) and 3 placed in -80 freezer, box Yohei (labeled “-80 V”)
Slow freezing solution made with the following amounts:
10 mL total solution with 1.5M DMSO and 0.2M Mannitol
DMSO - 1.09 mL
Mannitol - 0.4004 g
MBSH Water - 8.91 mL
With this 10 mL solution, 6 1.5 mL tubes were loaded with 1 mL each. 3 of these tubes were placed in freezer C, box 5 (labeled “CS”) and 3 placed in -80 freezer, box Yohei (labeled “-80 S”)
The remaining 4 mL from both the vitrification and slow freeze solutions were left out on the red tube rack
