Transformation was done on 5/25/21 on LB +AMP Plates made on 5/25/21 by HS. Transformation done by LC and HS.
KCNJ3-1 pGHE with a concentration between 899.28ng/uL and 1762ng/uL. DNA from Lane 3 on LMC052421B.
There is no bacterial growth indicating that we did not have a plasmid of KCNJ3 like we thought. This transformation was ran with 2 other positive controls which both grew indicating this was not an error in the transformation protocol.
Negative Control: KCNJ3-1 with a concentration of 69.24ng/uL. DNA from Lane 7 on LMC052421B.
Negative Plate looks good. There seems to be no colony growth which is expected for a linearized product.
KCNJ9 pGHE-2 with a concentration between 327.84ng/uL and 705.752ng/uL. DNA from Lane 10 on LMC052421A.
The plate indicated the DNA was circular and that transformation likely occurred. The colonies are too small and close together. This is probably due to the concentration of the DNA being too high.
Negative Control: KCNJ9-1B pGHE Linearized with a concentration of 39.06ng/uL. DNA from Lane 9 on LMC052421A.

OPRM1-1 pGHE with a concentration of 423.73ng/uL. DNA from Box 18.
The plate indicated the DNA was circular and that transformation likely occurred. The colonies are too small and close together. This is probably due to the concentration of the DNA being too high.
Negative Control OPRM1Purified Aliquot with a concentration of 16.30ng/uL to 25ng/uL. DNA from Lane 2 LMC052421A.
