HiFi DNA Assembly Subcloning is now being used in the Opioid Project to transfer the protein coding sequences of KCNJ3, KCNJ9, hOPRM1, GRK2, and ARRB2 to the pGHE plasmid. The source of the pGHE backbone will be the TAAR1_pGHE or B2AR_pGHE plasmid, coding sequences for the proteins of interest will be purchased from DNASU. Plasmid maps of our target products (including HiFi primers) are provided on the Opioid Project page at the bottom in the (.ape) format along with an updated HiFi primer word document with more advanced information. Completed projects and their methods can be found on the finished subcloning projects page. For the coding sequences of the proteins not yet complete, individual pages with step by step plans for their completion can be found on the unfinished subcloning projects page.
