pkk-mRuby3-TEV: Plate Two
While both plates had a couple of colonies that grew to a decent size, plate two had more colonies that were separated by some distance, rather than grouped together.
OPRM1: 1ng/uL
The 1ng plate contained a larger number of colonies that were of decent size and separated by distance. The 10ng plate had colonies that were larger, however, there were very few of them. This was due to improper pipetting during transformation. To ensure that the best DNA is obtained after maxi-prep, the 1ng was chosen.
KCNJ9: 1ng/uL
The 1ng plate had colonies that were of the correct size and separated by distance. The 10ng plate had more colonies so they competed for nutrients and did not grow large enough for a substantial inoculation and maxi-prep.
KCNJ3: 2B 10ng/uL
The 10ng plate had colonies that were of the correct size and separated by distance. The 1ng plate had more colonies to compete for nutrients so they did not grow large, despite the fact that the DNA used in the 1ng plate was of lower concentration. This has also been noted on the 5/27/21 transformation page.
Two colonies will be inoculated from each plate, for a total of eight maxi-preps.
