Lanes 2-5 were loaded with the usual 4 uL of MGB H2O, 1 uL of DNA, and 1 uL of DNA dye. Lane 6 was loaded with 0.25 uL of DNA and 4.75 uL of MBG H2O. Lane 7 was loaded with 0.5 uL of DNA and 4.5 uL of MBG H2O. The gel of lanes 2 and 4 was pierced during gel loading. Lanes 2-5 have a streak pattern which makes the actual band of DNA somewhat difficult to distinguish from the smear for Lanes 2, 4, and 5. Lane 6 showed no bands whatsoever. Lane 7 had a possible band that is indicated above, however, it is barely noticeable so it was marked to ensure that any DNA there that was shown was recorded with a concentration. Because of the lack of bands in lanes 6 and 7, two new aliquots of 20 uL of 200 ng/uL were made from the eGFP 3241.7 ng/uL and eGFP 6.112 ug/uL tubes. Those were run alongside the old aliquots of those tubes in the second gel below. All eGFP tubes found in freezer A, including the ones form lanes 6 and 7, were nanodropped to determine exact concentrations, and those can be found under the 6/17/21 Nanodrop results page.
All lanes were loaded with the usual 4 uL of MBG H2O, 1 uL of DNA, and 1 uL of DNA dye. The bands of all lanes are not very concentrated, except for the bottom band of lane 4. The top band of lane 4 was only noted because of the slight band that was found there, but the bottom band was used to determine the concentration of the second aliquot of eGFP 3241.7 ng/uL. Due to the lack of concentration for the bands on this gel and the two bands found in lane 4, the concentrations for the original aliquots were taken from the nanodrop results, and the 200 ng/uL concentration will be used as a reference for the new aliquots.
