Lesson: Microscopes and Biological Techniques

- How do biologists gather data?

This unit instructs students on the basic tools of lab biology which allow for the observation and collection of data to build our understanding of the workings of nature. Students will also conduct their own investigations, utilizing the proper tools and techniques to keep the laboratory a safe place.

    1. There are essentially three types of microscopes- know the differences in the way they work, how the images produced vary and/or are enhanced, and limits to magnification.

    2. Utilize a light microscope to make a variety of observations at different magnifications and demonstrate an understanding by preparing a wet mount slide and making adjustments to both resolution and magnification.

    3. What are some examples of other types of imaging used by biologists?

    4. Recognize that biologists use a wide variety of techniques to study life and be able to explain at least one of them.

- centrifugation

- gel electrophoresis

- chromatography

- labeling & fluorescence /micro-array (gene splicing)

- cell and tissue culture

- laboratory apparatus and tools

- PROBEWARE

Warm Ups

Microscope Chart - Students complete a review of the operation, uses and limitations of microscopes in the biology lab. #content

Imaging Tools - Students brainstorm a list of tools used by biologists to gather visual information/data

Notes

Handout - Microscopes - This can be covered in a demonstration prior to microscope. Most of the information matches up with the notes if the student is completing the assignment independently.

Independent Assignments & Activities

Web Assignment - Microscope Imaging - (Blank Document) Web Extension - Nikon Small World Competition Students examine how microscopes work and the types of images they can produce. Students search for interesting/intriguing images taken using microscopes and compose their own questions.#content #criticalthinking #community #collaboration #communication #creativity

Lab & Group Activities

Lab Activity - Microscopes

- Students complete the microscope handout, either as a class or as individuals using the notes, while also reading the instructions for proper preparation and viewing of specimens. Proper use and protocols are discussed and demonstrated prior to making observations. Students make at least 4 separate drawings of specimen observed through a microscope. They must record the sample and total mag. for each drawing. At least two separate magnifications must be used. One image must be from a slide/specimen they have prepared (swamp water/ pocket lint/ natural human vs. artificial hair vs. animal hair...)
