Part of the European Innovative Medicine Initiative research program "StemBANCC" focuses on the development of new toxicity tests, using induced pluripotent stem cells. Our lab contribution to StemBANCC consists mainly in defining and implementing mathematical models of the transport and cellular effects (mainly oxidative stress) of the tested molecules. Such models will then be used to extrapolate the results obtained in vitro to actual therapeutic dosing, in vivo. Our pharmacokinetic and systems biology models will be calibrated with experimental data obtained by the partners of StemBANCC. This PhD work will focus on the development of a library of SBML models for the main pathways of toxicity described in the scientific literature, Those models will be applied to the solving of scientific questions (e.g., on the control of oxidative stress, or of cell differentiation) posed by the definition of a toxicity test gallery using the induced pluripotent stem cells developed by StemBANCC.
Keywords: mathematical modelling, predictive toxicology, systems biology.
