ED-SCREEN is a 3-year French Project funded by the PNR EST by ANSES which will run from 1st April 2022 to 31th March 2025.
ED-SCREEN is focused on the development of a computational tool aimed at providing an insight into Endocrine Disruption modes of action for the Estrogen, Androgen, Thyroid and Steroidogenesis (EATS) pathways. Information on EATS mediated endocrine disruption will be gathered at two levels: a chemoinformatic analysis and a bioinformatic analysis. Moreover, information on basic toxicokinetics (TK) properties will also be provided. The analysis of these sources of information will enable decision makers to prioritize chemical lists as a function of hazard linked to EATS modalities and the TK profile of chemicals.
In the framework of this project, information about the disruption of EATS targets will be provided by QSAR models. Models will cover both nuclear receptor mediated effects as well as other potential modes of actions (e.g. TPO for thyroid disruption). QSAR models will also be developed for the prediction of oral adsorption and volume of distribution.
Archived (in vitro/in vivo) transcriptomics data for Endocrine Disruptors Chemicals (EDC) will be analyzed with innovative bioinformatics approaches such as gene co-expressed networks (WGCNA) and Qualitative Gene expression Activity Relationships (QGexAR) to define gene signature profiles associated with EDC as well as the transcriptomics space of EDC (in comparison to other chemicals).
The project will assist decision-makers by providing a top-down approach that starting from multidisciplinary data will summarize the potential risk of chemicals thanks to a unique index while visualizing detailed and multi-layered information.
