To prepare a mount of a leaf peel to show stomata and record the observations and draw pictures.
Rheo leaves (or) lily leaves (or) canna leaves, slides, coverslips, watch glass, water, glycerine, safranin, droppers, brush, etc.
Take a Rheo leaf and fold it at the centre.
With a sharp blade, gently pull out the lower epidermis of the leaf.
Transfer the peel to the water in the watch glass.
Add 2 drops of glycerine onto the slide and transfer the peel onto it.
Cover the specimen with a coverslip.
Observe under the microscope.
The cells are irregular or rectangular in shape. They are lower epidermal cells.
At certain places, stomata are seen.
Stomata consist of 2 guard cells, followed by subsidiary cells.
Some opened and closed stomata are seen.
should not be allowed to dry.
Avoid air bubbles while covering with a cover slip.
Use fresh leaves to see open stomata.
Take out only the lower epidermal to observe stomata.
