RELEVANT LEARNING OUTCOME:
1 The Cells and Biomolecules of Life
(b) For practical assessment, candidates may be required to operate a light microscope, mount slides and use an eyepiece graticule and a stage micrometer.
The light microscope and the electron microscope are used to observe cells and their organelles, as well as non-living samples, such as the starch grains (viewed through a light microscope) that you see in the banner above. Pictures can be taken of the images and these are called micrographs.
👁️🗨️Watch the video to see how a light microscope works and how it is used. Do note the care instructions from 7:22 onwards that will be very useful for your practical sessions.
🤔Think about this: To be begin viewing the slide using the scanning objective lens (magnification of 4x), should the stage be at its lowest or highest position?
Getting a specimen into focus
Calibrating the eyepiece graticule that allows for
-- measuring the specimen
-- calculating the magnification of a drawing of the specimen
Making drawings of the specimen (planned or detailed)
