Deliver these resources to Asseliander. You do not need to hand in all resources at once. It is possible to hand in portions of it and the progress of the quest you can see in your Attributes under Quests.
Afterwards you need to activate the Braziers in Tentacler Valley, Daigon Islands Scolopendra Coast, Silent Cove and Azure Lagoon. In each location you will have to fight a monster:
Custodian of Silent Cove, 2117 health, hits for 130-180, casts same effects as Serpagons
Custodian of Azure Lagoon, 6422 health, hits for 125-180, casts same effects as Scolopendras
Custodian of Scolopendra Coast, 6894 health, hits for 130-185, casts same effects as Pearly Scolopendras.
Custodian of Daigon Islands, 5072 health, hits for 185-245, casts ame effects as Daigon
Custodian of Tentacler Valley, 810 health, casts same effects as Tentaclers
As a final fight you need to kill Poisedopus Shadow. For this fight you get an extra spell that you have to equip to your spells list before the fight. Additionally a lot of concentration is required (thus Elf Spy Cloak or better is recommended). You do not need to bring manas to the fight as you will receive Gloomslayer Gift for the fight.
Recommended slots: Giant, Spirit, Atshi, 3 slots of red Scrolls of Retribution and the rest lifes. Possibly add a celebration punch.
Recommended fight tactics: When he reaches ~70% health he will start to hit twice per turn, at ~30% he will start to hit 3x per turn and at ~15% he will start to hit 4x per turn. At the first threshold is a good point to start with scrolls of retribution, keep him under the effect at all times. When he starts to hit three times per turn start using retribution arrows. If you are in the possession of a Clan Totem on the last threshold is a good point in time to use.

Twilight Runes
By using a Twilight Rune, a slot gains an illusory copy. This copy can be filled like a normal belt slot. During the fight it's possible to swith between the different copies of the slot before any of the items are used. The contents of the other copies of that slot can then not be used anymore.
This feature is particularly useful when several options of potions or scrolls might be needed in a fight. An example is the random superbeing you get attacked by when using a Superbeing Card Pack.

Four slots can be gained from using Blinding Twilight Runes with the first one costing 1 Rune. Each subsequent use will double the cost. So the second slot gained from them will cost 2 Runes, the third will cost 4 Runes and the last slot will cost 8 Runes.
Using one rune will get you an additional pocket in your first belt slot. It will look like this:
