Genetics Call 2015-10-26
Post date: Oct 26, 2015 3:34:51 PM
Case-control, moderate to severe, severe; lung function - updates
Many people not able to make the call; email sent to at least one member of each cohort.
Would like to confirm additional authors -- guidelines up to 4 per cohort
QC: will plan to send around to review unless objections. Outstanding issues are Lovelace imputation; LHS also plans to contribute a FEV1 analysis.
Meta-analysis: assuming QC is reasonable, should have independent analysis confirmation of at least the top associations
Additional analyses e.g. set/pathway, eQTL lookups, cross-ethnicity.
Replication vs additional power for discovery: additional cohorts genotyped by Ed Silverman / Channing including Early-Onset COPD, ICGN, TCGS.
Discussed with Martin Tobin: plan to take top associations and lookup for COPD in BiLeve
Emphysema analysis
Still in process of deciding on analytic issues, which are myriad
Edwin Van Beek mentioned importance of looking at protocols: e.g. protocols that were close to COPDGene, SPIROMICS, ?MESA vs other low dose. Would be instructive to compare distribution of %emphysema within a specific GOLD classification, and take a look at sample of individual histograms.
