Vaccine Construction

For the construction of putative vaccine candidate sequences, high scoring B-cell, CTL and HTL epitopes were used which were ligated with adjuvants such as Beta-defensin (Accession ID: AGV15514.1) to enhance the immunogenicity of the synthetic vaccine sequences.

Illustrative diagram of our best multi-epitope vaccine construct for Chagas disease. The vaccine construct consists of 27 epitope sequences, each sequence being 9 amino-acid long, belonging to CTL, HTL and BCL epitopes of our 9 filtered proteins. Beta-defensin (Light purple) has been used as the adjuvant and is linked to the top CTL epitope (light pink) using an EAAAK (maroon) linker. The other CTL epitopes are linked to each other using the GGGS (violet) linkers. The last CTL epitope and the first HTL epitope (blue) as well as the other HTL epitopes are connected through the AAY (sky blue) linker. The last HTL epitope and the first BCL epitope (yellow) as well as the other BCL epitopes are connected through the KK (purple) linker. An EAAAK (maroon) linker has been added at the end of the sequence for increasing the stability of the vaccine construct.
