Introduction :
This lab inquiry will include the use of the Edvotek Megacycler, a thermocycler which produces multiple copies of a target DNA molecule. The results will then be analyzed by gel electrophoresis. Students are expected to know and understand the details of the process.
Lab Questions :
1. What is the function of the DNA Polymerase required for PCR-based DNA amplification? What type of substance is it?
2. Why are dXTPs, like dATP,dGTP; required for the PCR reaction? What material in this lab procedure contains these molecules?
3. What are possible reasons for obtaining fainter bands besides the main PCR product in the gel electrophoresis analysis?
4. What is the function of the Ethidium Bromide?
5. What is the purpose of the 200 base pair DNA ladder in the gel electrophoresis ?
6. What tool is needed to view a gel which has been treated with Ethidium Bromide?
7. Define the following terms :
a. PCR
b. Primer
c. Taq DNA polymerase
d. What is the source of Taq DNA polymerase
8. Lab Procedures : Describe why temperatures rise and fall in the PCR procedure.
