Proteins (Practical)

About the Course

Dear learners welcome to the practical sessions of proteins. The lab exercises and experiments provided in this manual are based on the units that you have studied in BBCCT-105. The concepts that you have studied in theory paper of proteins will be performed and experienced during this lab course.

Objectives

The broad objectives of this lab course for how to enable you to:

  • Know the different types of principles and procedures available are the estimation of proteins

  • Impart the skills of isolation of protein

  • Enrich with protein fractionation procedure

  • Distinguish between extraction, isolation and separation techniques,

  • Perform separation of proteins using electrophoresis;

This lab course is off 2 Credits and consists of eight laboratory experiments. The experiments are designed in such a way that you will be e performing estimation of proteins by using UV absorbance and Biuret method. Also the other two methods popularly known as Lowry method and Bradford method have been included.

Since you have studied about extraction isolation and analysis of proteins in the theory paper you will be e performing the isolation of casein from milk using Isoelectric pH method. In addition to this you will be performing ammonium sulphate fractionation of serum proteins through which you will be learning how to concentrate the protein sample after extraction.

The last two experiments designed in such a way that, they will provide you hands on experience on how to separate serum proteins using paper electrophoresis and SDS-Page.
