Fluorescence Activated Cell Sorting

The Facility is currently equipped with two state of the art high speed cell sorters on the St James's University Hospital Campus UoL. All our systems are capable of sorting into FACS Tubes and into a variety of collection plates with index sorting capabilities.

The Influx is our high end digital cell sorter housed in a CAT 2 hood allowing for the sorting of unscreened patient samples and other more high risk samples including transient transduced cell material (the facility will require a copy of the risk assessment of material prior to sorting). This system is run as a service by facility support staff due to the complexity of the system. Bookings must by made using the QReserve Online Calendar System. Please confirm staff and system availability and email medlfacs@leeds.ac.uk to confirm the sorter booking with staff.

System capabilities:

  • 5 laser system (355nm 100mW, 405nm 100mW, 488nm 200mW, 561nm 70 mW & 640nm 120mW)
  • 18 fluorescent parameters alongside Forward scatter (FSC) & Side Scatter (SSC)
  • 2,4 and 6 way simultaneous population sorting into 5ml FACS tube sorting output
  • Temperature control on sample station and collection vessel
  • Plate sorting capabilities into 6, 24, 48, 96 and 384 well plates including index sorting single cell capabilities for downstream next generation sequencing work
  • 70µm, 86µm 100µm & 140µm nozzle attachments with sorting rates up 70,000 cells/second
  • Enrichment, Purity and Single Cell sorting mask modes for highest possible sorting and maximal yield outputs
  • Up to 32 sort regions possible
  • Collection tube holders include sizes from microtubes, to 12 x 75-mm tubes, to 15-mL tubes
  • CAT2 Hood and Buffalo Aerosol Extraction Unit for high level containment of biomaterials
  • Sortware acquisition/analysis software

The FACS Melody is our newest digital cell sorter. This easy to use system can be run by researchers (training is provided by support staff) or by facility support staff (dependent on staff availability through QReserve). For training requests or to arrange for staff to operate the cell sort please confirm staff and system availability and email medlfacs@leeds.ac.uk to confirm the sorter booking with staff.

  • 2 way simultaneous population sorting into 1.5ml, 2ml and 5ml tube sorting output

  • Plate sorting capabilities into 6, 24, 48, 96 and 384 well plates including index sorting single cell capabilities for downstream next generation sequencing work
  • 100µm nozzle attachment with sorting rates up 40,000 cells/second
  • Enrichment, Purity and Single Cell sorting mask modes for highest possible sorting and maximal yield outputs
  • Up to 32 sort regions possible
  • Buffalo Aerosol Extraction Unit for improved level containment of biomaterial aerosols
  • Chorus acquisition/analysis Software
