Your first goal is to complete Adventure mode as far as possible.

Adventure Mode is the main way to level up your dragons, which is utterly imperative to proceeding in the game.
Doing Adventure mode fulfils Master's Path Quest rewards, and you can pick up other resources such as gold and diamonds from it, which will help you progress greatly in other areas!
Once you reach Hard mode, you can pick up 4-star runes!
The later chapters of Hard mode gives out 4-star runes of a specific slot and colour, which is a huge help in runing up your dragons.
At first, just focus on completing Adventure mode on both Normal and Hard. Then you can proceed to the Nightmare Dungeon, as you will usually need good runes to tackle Hell mode
Don't forget that you can borrow a friend's dragon to help you on your Adventure!
So now you're in the middle of you Adventure, and you realise your dragon inventory is getting a bit cramped. Besides which, you're finding the Adventure stages increasingly harder. What can you do to avoid hitting a wall?
Do the following, in rough order that you should do them:
The great bane of every tamer's life: runes. You will need dozens upon hundreds of these, and unfortunately, the need to grind runes never stops.
When you are first starting out, however, don't worry about them too much!
Just equip what runes you have to your dragons at first. They will likely be 1-star to 3-star runes, so you do not need to enhance them - though you can do so up to +6 or +9 if you find that you need just a little more boost to your dragon's power. (Note that enhancing 1-star to 3-star runes gives such little returns that it is usually not worth it.)
In Adventure Hard mode, you can begin to farm for 4-star runes. It is ideal to farm for the desirable colour and main stat at this stage. I advise you not to enhance them to beyond +12, however. Enhancing a rune up to +15 is expensive, and is not worth it for runes you will easily replace.
After you get 4-star runes on your main party, you should hopefully be ready to tackle the Nightmare Dungeon.
Complete Nightmare Dungeon levels 1 to 6 purely to progress to the later levels. They have a chance of giving out 3-star or lower runes, which are not worth your attention.
Starting from Nightmare Dungeon level 7, you can obtain 4-star to 6-star runes. Levels 7 and above are the ones you will want to farm. See the linked page on Nightmare Dungeon for more information about this area.
Once you have optimal runes of 5-star or higher grade runes, you may be ready to begin working on the Rune Guardian dungeon.
An aspect of the game that you are quickly introduced to, but is worth mentioning nonetheless.
Should you spend all your initial diamonds on summons? Yes, absolutely. When you begin, you need a lot of good dragons, and you also need a lot of bad dragons that you can use to rank up your better dragons and make them stronger.
One thing I will warn you about is to be careful that you do not oversummon. If you have more dragons in your inventory than you have inventory space, you will not be able to go on Adventures, Explore, or most other content until you have either expanded your inventory, or got rid of the excess dragons.
A good way to get around this is to make sure your existing dragons are already max levelled, so that you can use new fodder dragons to rank up your existing dragons immediately. This way, you can immediately get rid of excess dragons in a useful way.

When you first start out, Colosseum seems an impossible consideration. How on Earth are you meant to compete when your main team are not even Adult dragons yet?
To which I say, you are absolutely right, but there is no need to neglect Colosseum! It gives out great rewards, so whenever you have refilled to 10 tickets (you receive 1 ticket refresh every hour), set the game to repeat, if you must, and let the game use up your tickets on auto.
As you get more and better dragons, and perhaps even a better feel for the competitive scene, you may wish to begin investing in Colosseum as well. They are one of the only ways to interact with other players (and even then ... you don't really interact with anyone, as the team is controlled by AI ...), which makes it a fresh, ever-changing area of content as people come up with new teams and strategies every day.
