Tuesday 2/20/2024
Today was our first real test for data. We made all of the dilutions and streaked the plates. We then labeled the plates and marked the spots to put our discs. We then dipped one of the paper plates in the dilutions and put them on the correlating plate.
Wednesday 2/21/2024
Today we analyzed our plates from Tuesday. Our chosen analysis method was to measure the zone of inhibition (ZOI) produced by the disc mixed with the dilution. After looking at the plates we realized that all had a ZOI of zero. This means that hot sauce does not affect the bacteria or that our hot sauce wasn't strong enough. To test this theory we planned to use a stronger hot sauce in the next trial.
Thursday 2/22/2024
Today we tested our theory from yesterday and used a two-million Scoville hot sauce. We prepared the plates the same as we did for the Tabasco but instead using the Scorpion Hot sauce. After that, we put the plates into the incubator to incubate for one day.
Friday 2/23/2024
Today we analyzed our results from yesterday. Once again we saw that there was a ZOI of zero. After this realization we started to question if our bacteria was the variable that could not be affected so we decided to try three new bacteria; B. Coagulans, S. epidermidis, and L. AcidophilusÂ
