The first two Challenge Levels require 5 and 15 Challenge Points, respectively.
From Challenge Level 3 onwards, they require (10 × Challenge Level) Points to progress to the next level and gain the unlocks.

Cards in starting deck have the Annoying sigil.
Annoying: The creature opposing a card bearing this sigil gains 1 Power.
The sigil is added to the innate sigils on the card.
If you had already beaten the Weak Start challenge before the update, Annoying Starters would be checked off as completed.
If you have an Opossum and/or Ring Worm in your starting deck because of the reset mechanic, they will have the Annoying sigil as well.
Bugs
The Annoying sigils are treated as added sigils in Goobert’s node and can be changed, despite displaying as innate.
V0.22: FIXED
Tadpole, Kingfisher, Dire Wolf Pup are not affected by Annoying Starters, since they already has two innate sigils.

Your squirrels have the Waterborne sigil.
Squirrels gain Waterborne and become a new card, Aquasquirrel.
Bottled squirrels are unaffected.

45 Challenge Points
Grizzlies appear during the first 3 boss battles.
Spawns a full board of 8 Fucking Bears 8 Mighty Leap Grizzly Bears at the start of the second phase for boss battles (except Leshy.)
The bears' appearance disables the second phase boss gimmicks (Prospector does not board wipe, Angler does not place Bait Buckets, Trader does not give you a pelt or allow trading.)
This is the same as the bears that appear in the base game when fighting the Angler/Trapper on your first/second run, respectively, except the game does not display the “Too Fast, Too Soon” message.
Changes
Increased Challenge Point value from 45 to 50.
Unlocked by beating Challenge Level 12.

Leshy is replaced as the final boss of the run. Heave. Ho.
Royal replaces Leshy as the final battle.
Unlocked by beating Challenge Level 8. Both instances are unlocked.
10 Challenge Points
Cards in your starting deck have 1 less health.
Cards are renamed to Weak [Card Name] unless that card already had 1 health, in which case it is unaffected. If a weak card is used in Mycologist Event, it retains the “Weak” moniker in its name.
If a Weak card is given Fledgling, it will not change its name from Weak [card name] to Elder [card name] / Elder Weak [card name] when it grows.
