The Use of Cell and Viral Mutagenesis to Improve the Detection and Study of DNA Viruses in Fish

ABSTRACT

When it comes to food security, aquaculture is one of the most important sources of nutrition. To optimize aquaculture operations, we must improve our ability to study and diagnose important infectious diseases that affect critical species. A group of understudied, but important pathogens are DNA viruses such as herpesviruses, iridoviruses, and poxviruses. Viruses in these families are known to significantly impact various species of fish vital to several aquaculture sectors. This 30-minute work-in-progress seminar will focus on the limitations to the study of certain viruses and how we can use viral or cell mutagenesis to improve the study and diagnosis of these pathogens in fish. Among the discussion topics is 1) the generation of immunocompromised fish cell lines that make possible in vitro isolation and propagation of currently unculturable DNA viruses like Carp Edema Virus (CEV); and 2) the generation of viral mutants via Bacteria Artificial Chromosomes for investigation of temporal viral distribution within the host, particularly as it pertains to latency establishment in DNA viruses like Acipenserid herpesvirus 2 (AciHV-2). A summary of each virus and its importance in the aquaculture setting will be reviewed.
