Please submit a paper copy for the Lab Report:
A. Materials :
1. Plate culture of Armillariella mellea
2. Sterile scalpel
3. One Bread Crumb Agar Plate
B. Procedure:
1. Use the sterile scalpel to cut 1 - cm cubes from the growing edge of the culture.
2. Invert the cube in the middle of a bread crumb agar plate.
NOTE : Avoid touching the cutting edge of the scalpel and the inside surface of the plate to avoid contamination.
3.Use a marker to write your name on the plate in a small area.
4.Incubate the plate in a dark room at 25 Celcius.
5. New hyphal growth will be observed in one week. Rhizomorphs form in 10-14 days, and bioluminescence begins .
6. Observe and record observations and pictures once per week ( or more) for four weeks.
a. Observe the plate in the light and record a photo and observations.
b. Observe the plate in darkness, and then record observations. It takes about 5 minutes for your eyes to adjust enough to see the low intensity, pale glow. Record the color.
c. Record Observations:
Including measurements, descriptions, a scientific note : date, time
Four Photos over four weeks in a document . Labels are required.
