Populate your content, bearing in mind the notation of what needs to be in each of the marked columns so it goes to the correct place when folded. Don't forget to remove the textboxes with that notation.
Before printing, right click on the grey frames around the columns and the margins and choose Unlock from the pop-up menu. This will make them available. Click on them and press the Delete key on the keyboard. There should only be two little pale grey lines at the top and the bottom end of the middle of the inside spread area of the leaflet. They are used when folding, so keep them there.
When printing, make sure that you select to print double sided, flipped on short edge. You need to choose to print Full Page Slides and make sure that you untick the option "Scale to Fit Paper" from the same menu.

The two light grey lines that you will see on the printed leaflet should be used to fold the cover to just touch them. Once that outer fold is nice and firm, open the leaflet again and now fold the inner piece so that the gray lines are exactly in the fold itself. That inner page of the leaflet is designed to be 2mm narrower than the other two columns to allow for an easy fold and for it to fit nicely in the leaflet.
