Make the slide
First of all, we need to make a chamber. We use this chamber to hold the fluid with Holliday junction. A chamber consist of two pieces of glass, separated by double sided tape. Since you need to flush the channel multiple times, you use a glass slide with holes drilled into it. The coverslip is cleaned with KOH before you receive it.
- Get a microscope slide with holes
- Lay the slide on a tissue, so it won’t get damaged
- Now cut multiple pieces of double sided tape, and lay them along short side of the substrate, in between the holes
- Take a cover glass from the box you get from your TA
- Place the cover glass in the middle of the slide
- Cut the double sided tape along the cover glass and remove it.
- Seal the chamber with epoxy
You now have created multiple little chambers between the slide and the cover glass. This chamber will hold around 100 microliter.
