Outline

Measure the state of the Holliday junction

When Cy3 is close to Cy5 (isoII), Cy3 donates its energy to Cy5, and Cy5 emits this energy in the form of light . In isoI, Cy3 can't donate its energy to Cy5, and thus Cy3 will emit light by itself. The microscopy setup is capable of splitting light of different spectrum into two images on the camera (Cy3 vs Cy5).

Figure 1. Holliday junction (a model system to study).

You will receive Holliday junctions with the Cy5 and Cy3 already attached.

Goal

You receive three samples, with three Holliday junctions. By measuring the dwelltimes, you have to determine which sample contains which junction.
