We seek to identify pathways or genes which when mutated bring about premature ageing in humans. Inactivating mutations in PYCR1, P5CS, MTX2, TAPT1 or RAF1 lead to such segmental progeroid diseases in humans. These may lead the way for therapeutic interventions. For instance, maintaining the activity of these pathways could increase lifespan/healthspan in the context of chronological ageing. Conversely we may want to specifically block these genes in the context of tumorigenesis to prevent cancer growth.
