According to Mary Shelley's canon, Victor's mother Caroline died of scarlet fever caught from Elizabeth, but in Branagh's film, she died giving birth to Victor's younger brother William. In the book, she had a chance to speak to both Victor and Elizabeth, expressing her love and hope for reunion in an afterlife. In the movie, she died without getting to properly say goodbye and neither Victor nor Elizabeth were present. She insisted the baby was saved instead of her.
In both films, their loving home was filled with joy and happiness, but it quickly faded after her death and everyone fell into depression.
Caroline's death sent Victor over the edge and his inspiration quickly spiraled into obsession. It became a fixation with immortality that led to his madness and moral decay.
