MB21D1, a cadherin-binding molecule, was identified as a signaling regulator for KRAS and PI3K pathways. Anti- MB21D2 therapy was found effective to treat cancers with active KRAS or PI3K signaling networks.

Heterophilic interactions between melanocyte and keratinocyte maintain skin homeostasis. UVinduced CDH mutations cause contact control loss from keratinocyte and trigger melanomagenesis. During metastasis, mutation-derived Neo-Ags boost T-lymphocytes and activate anti-cancer immunity, which in a reverse way is beneficial for immunotherapy.
