Removed petri plate of mother colony from incubator and took pictures

• • Analyzed using ImageJ Software Colony Blob Count Tool

  • There were 67 colonies and an average area of 21182.97

Calibrated turbidimeter with distilled water, shows 0.357 NTU error so subtracted 0.357 NTU from raw data

Put 3 mL Luria broth into turbidimeter cuvette, inoculated with bacteria, recorded 3 turbidity measurements and took the average, transfer solution to corresponding sample vial

• • Repeat for Samples 44-60

Incubated vials at corresponding temperature

• • Will be ready to remove from incubator on 2/18/21
