Removed Group B PET (Samples 31-60) from UVC light
Using forceps, dipped each PET strip into beakers with distilled water then ethanol, dried with Kimwipe, recorded mass in Excel
Placed PET strip to corresponding sample vial
Calibrated turbidimeter with distilled water, shows 0.226 NTU error so subtracted 0.226 NTU from raw data
Put 3 mL Luria broth into turbidimeter cuvette, inoculated with bacteria, recorded 3 turbidity measurements and took the average, transfer solution to corresponding sample vial
Repeat for Samples 6-10, 16-20, 26-43
Will be ready to remove from incubator on 2/12/21
Samples 31 and 32 have extreme turbidity readings because it was a different bottle of Luria broth and it might be expired
Only got to Sample 43 because agar plate of mother colony was pierced and couldn’t inoculate vials with enough bacteria for remaining 17 vials
Incubated vials at corresponding temperature
Inoculated new agar plate with original rehydrated culture vial in liquid suspension
Incubated at 30℃ and will remove on 1/19/21 for 4.5 day incubation period
Will inoculate remaining vials and measure turbidity on 1/19/21, don’t need to re mass PET strips because they are already in corresponding sterile vials
