Got 144 5mL glass vials and 3 incubators available for me to use
Removed petri plate of mother colony from incubator and took pictures
Analyzed using ImageJ Software Colony Blob Count Tool
There were 67 colonies and an average area of 21182.97
Rinsed 30 PET strips (Group A, Samples 1-30) with distilled water and ethanol over a sieve
Dried with a Kimwipe and massed in Excel to the nearest ten thousandth
Had to trim and re mass samples 1 and 2 to fit into the vials
Prepped 10 vials with 3 mL of Luria broth but then realized its more efficient to put broth and inoculate bacteria directly in the turbidimeter cuvette
Calibrated turbidimeter with distilled water, shows 0.457 NTU error so subtracted 0.457 NTU from raw data
Measured turbidity 3 times for each sample and took the average of each
In each vial, put 3 mL of Luria broth cuvette, inoculated with bacteria, measured turbidity, and transferred to sample vial with PET strip
PET strip corresponding with sample number of vial
Only got 15 vials done so did 5 from each group (Group 1A, 2A, 3A)
Hopefully I have enough Luria broth for rest of samples
Incubated samples at temperature corresponding to each group, will remove on 2/10/21
Vials too small to stand up in incubator had to place in a beaker then in incubator
Labelled all vials with wax marker
