Invadopodia colocalization macro

This macro is a handy tool to count the number of invadopodia in a cell, defined as the colocalized dots of two invadopodia markers (e.g. Tks5 and Cortactin). The macro converts multi-point ROIs, representing putative invadopodia, into colored circular ROIs. The steps for using this macro to count invadopodia in a MDA-MB-231 cell stained with invadopodia markers, Tks5 and Cortactin, are described below:

1 : In the Tks5 image, run Process/Find Maxima… to find all the putative invadopodia dots.

2 : Transfer points from the Tks5 image to the Cortactin image (Ctrl+Shift+E).

3, 4 : run Invadopodia colocalization macro to convert multi-point ROIs to colored circle ROIs

5 : Analyze images visually for colocalization and count dots containing both Tks5 and Cortactin as invadopodia.

Note: Tks5 dots in the bottom right (marked by red arrows) are not invadopodia because they do not contain Cortactin
