Last time we checked in, we had begun our CBD pretrials to determine the dosage of CBD. Unfortunately, 4/5 flies died in the process. We believe that by sprinkling the CBD on top of the media, we had created a seal over the food, and our flies starved. Luckily, our wonderful mentors suggested that we make 3/4 of the regular media first and let it solidify into a gel. Then, we make the last 1/4 of the media mixed with CBD and yeast. This way, the CBD will be incorporated into the media, but it will not sink to the bottom of the vial. We've redone our CBD pretrial, and 4/5 of our flies survived! We'll continue to use this CBD in our experimental trials.
We started on our first real set of trials by filming 30 of the no stress flies in our square petri plates. All was going well, and we finished these trials on time. Unfortunately, when we tried to upload these videos to the computer, it erased 17 of our 30 videos, but we kept our heads high and we have refilmed 10 of the 17 lost flies! We are continuing to refilm and age these flies, and, on November 13, we will finish filming our no stress control group flies. While this has been going on, we also began to film our first group of 10 stress, no treatment flies. We filmed them on November 4, and on November 5, we separated our next group of 10 flies into solitary confinement to be filmed on November 12. We are extremely excited to be finally moving into our experimental trials, and just by looking at our stress, no treatment, flies they seem to be exhibiting the wall walking behavior!
Being parents to hundreds of flies is no easy task. We constantly need to make new fly vials to keep our flies happy and healthy. On average, we make 4-5 vials a day (20 vials a week)! To make the perfect habitat, we mix dry fruit fly media with water and yeast. We wait for the mixture to solidify into a gel, and transfer our flies. As you can see in the gif, we use a funnel to transfer flies from one vial to another. We try to cap the vials with foam stopper before the flies gather their senses and escape. Unfortunately, some flies do escape, so we have to make an apple cider vinegar- hand soap concoction to trap them. Just another day in the life of a biotech student!
