Amnis ImageStreamX

The Advanced Flow Cytometry Platform offers users the Amnis ImageStream Mark II (ISX) which combines the quantitative power and speed of Flow Cytometry with high content image analysis. It quantifies both the intensity and the location of fluorescent probes and can analyse highly heterogeneous samples and rare cell subpopulations at a rate of up to 5000 cells per second.

The ISX provides new insights into the co-localisation of proteins, cell-cell interactions, internalisation, targeted immunotherapy, parasitology or cell cycle/ cell death related experiments. The technology is furthermore well suited for the study of extracellular vesicles such as exosomes.

ISX configuration

Our ImageStream MarkII is equipped with 2 cameras providing 12 image channels and 4 co-linear excitation lasers (405 nm, 488 nm, 560 nm, and 642 nm) as well as MultiMag (20x/ 40x/ 60x) and EDF (extended depth of field). Cellular imagery is split into its component colours through a unique spectral decomposition element. The result is a bright field image, a dark field image, and multiple fluorescence images of every cell. The ImageStream MKII uses the latest versions of INSPIRE software for acquisition and IDEAS software for analysis. The table below allows for an easy overview of possible fluorochrome combinations.

Booking

The analysers are available self-service only, 24 hours a day, 7 days a week for use by investigators except during weekly maintenance and repair periods.

Before an investigator can book and use the equipment, they must register with the Flow Core and attend mandatory Basic Training, in order to be eligible to book ImageStream training. This is because operating the ISX and designing experiments can be complex and requires a knowledge of fluorescent flow cytometry. Furthermore the necessary facility induction is only available with Basic Training. ImageStream training can be taken at any point after Basic Training and throughout the year, see our training dates.
