Conducted a preliminary experiment to determine the growth rate, seeding density, and optimal metabolite concentrations for C2C12 cells. Plated triplicates of four metabolite concentrations on a 12 well plate ranging from an upper limit of 1mM serine down to a lower limit of 0mM serine in C2C12 media. Imaging and counts taken at day 0, day 1, and day 4 time points.
based on imaging of four cell conditions at day 1 and day 4 time-points:
0.1mM ser best for survival
0.4 mM ser best for max growth rate
Determined growth rate to be 50,000 cells/hr within a 4 day experimental period.
Growth Rate = 1.14 1/day
Ideal seeding density of 5.0 x10^3 cells/cm^2
