Wet Mount
"Wet Mount" is a technique scientists use to observe moving cells under a microscope to get a clearer image.
Materials Needed:
1. Object of Experiment 6. Beaker with Water
2. Microscope 7. Dropper
3. Glass Slide 8. Knife
4. Cover Slip
5. Forceps
Steps of How to Prepare a Wet Mount Slide using an Onion:
Get a glass slide and cover slip for yourself and make sure they are both thoroughly washed and dried.
Remove a single layer of epidermal cells from the inner (concave) side of the scale leaf (The thinner the better).
Place the single layer of onion cell epithelium on a glass slide. Make sure that you do not fold it over or wrinkle it.
Place 3 drops of water on your onion tissue.
Put the cover slip on the tissue and gently tap out any air bubbles using the forceps.
Observe the cells under 10x with the diaphragm wide open. Slowly reduce the light intensity by closing the diaphragm, and observe the image.
