What is FXR?

Farnesoid X receptor (FXR) is a nuclear receptor that is encoded by the NR1H4 gene.

What is the function of FXR?

FXR is highly expressed in the liver and intestine. Bile acids are natural ligands for FXR. Similar to other nuclear receptors, FXR binds to its ligand and translocates to the cell nucleus. As binding to hormone response element on target DNA, FXR regulates its target gene transcription.

FXR regulates hepatic bile acid synthesis, gluconeogenesis, and lipogenesis etc.

What is Obeticholic acid?

Obeticholic acid is a semi-synthetic bile acid analogue. It has been FDA approved for the treatment of primary biliary cholangitis.

Currently, obeticholic acid is under global clinical trial phase III for the treatment of NASH.
