Procedure for Image Analysis (for photographs of petri dish, star photos, or any coloured maps) - Set the camera on the ‘Close-up' mode.
- Take a clear picture of the plate.
- Name and save the image as Image1.
- Click File -> Open -> Image1
- Change image to 8 bit by selecting Image -> Type -> 8 bit
- Adjust Threshold by selecting Image -> Adjust -> Threshold
- Adjust threshold settings so that just the colonies are highlighted in red.
- Click Apply. This will turn the image into a black and white binary image.
- Apply Watershed by selecting Process -> Binary -> Watershed. This will separate out colonies that are touching each other so that they will be counted separately.
- Count Colonies by drawing a circle around the colonies to be counted.
- Select Analyze -> Analyze Particles. Make sure the Display results, Clear results, and Summarize boxes are checked. Click OK.
- The summary panel will display the %Area.
- Repeat steps 1 to 12 for the other petri dishes.
