LC-QQQ-MS/MS

Targeted MS is important mainly because of the significant advantages of great selectivity and excellent quantitation. The most commonly used instrument for targeted metabolomics is the liquid chromatography triple quadrupole (LC-QQQ) mass spectrometer operating under multiple reaction monitoring (MRM) mode. Large targeted MS assays, covering ~280 metabolites in many metabolic pathways (such as glycolysis and the TCA cycle), can be constructed for metabolomics studies. Our own assay of this type has been successfully used in a growing number of studies, including metabolic reprogramming during Myc-regulated tumorigenesis, cardiac metabolic shifts induced by rapamycin, metabolic characterization of mammalian skeletal muscles, and metabolome regulation during naive-to-primed human embryonic stem cell transition.
