Questions for Matt on 2/23
Present Media from Sat test
- Needle test
- Glass capillary tubes
Dilution ratios recorded of 3:1 and 5:1 (show pics)
Let Matt observe beads start to gravitate toward needle
We came upon a lot of questions while trying to spread the beads into the wells:
- What's the best way to get a good distribution IN wells, and not around them?
- How much water do we ideally want on the slide?
- Is it possible to have water in JUST the wells and nowhere else on the slide?
- Should we care about a big water droplet?
What conclusions can we draw from these images?
For specific feedback:
- Say what we THINK we've learned
- How do you think we can pick up cells (or cell) specifically out of the well?
- Does this seem like good progress?
- What would you like to see by next week?
Glass needles:
- Present needle failures
- Ask for his impression of viability
- Use this info to decide on micro-injector order
