This instrument is very easy to use. Follow the instructions below to obtain an absorbance spectrum.
If you are the first person to use the instrument on a given day, you need to take a background spectrum:
Log into the computer.
Open LoggerPro (the spectrometer will automatically turn on).
Take a background spectrum:
Choose Experiment > Calibrate > Spectrometer. Wait for the spectrometer to warm up.
Place your blank sample in the spectrometer (usually DI water in a cuvette).
Make sure ridged sides of the cuvette are not in the light's path .
Press Finish Calibration.
Press OK.
If LoggerPro is already open, you can assume that a background spectrum has been taken.
Once calibration is complete, you can go ahead and take your absorption spectrum:
Have all of your samples ready.
If data is on the screen, select File > New, and do not save previous.
Place your sample in the spectrometer; make sure ridged sides of cuvette are not in light's path.
Press the Collect button to take spectrum.
Once the spectrum is visible on the screen, press the Stop button.
If you want to take the spectrum of another sample, repeat steps 3-5.
If you need to know the peak wavelength and/or amplitude, press the “x =” icon and copy down the appropriate numbers.
At the end of the day, turn the spectrometer off by closing LoggerPro
