It is believed that he was with the Pilgrims in Leiden, Holland. His wife's name is not truly known but has been referred to in various books to be Ann. Edward died in the died between 11 January and 10 April of 1621. His wife passed away after 11 January 1621. Samuel survived and Matthew came over before 1640.

The question of the paternity of Matthew Fuller was examined exhaustively by Bruce C. MacGunnigle, Robert M. Sherman and Robert S. Wakefield in 1986, and they came to the conclusion that Matthew was a son of EDWARD FULLER [ TAG 61:194-99]. Bruce C. MacGunnigle has published the definitive treatment of Edward Fuller and his descendants in the fourth volume of the Mayflower Society's Five Generations Project. " He was the oldest son of Pilgrim Edward Fuller and did not make the passage with his parents and younger brother, Samuel. When he arrived in New England, he was already married and a parent.
In April of 1642, ten acres of land was granted to him in Plymouth. Also in 1642 he served as juryman and was made a freeman. He was sworn and admitted on 7 June 1653. In 1643 he was appointed sergeant in the military company under Captain Myles Standish. Around 1650 he moved to Barnstable, Barnstable, MA, in 1653 he was a representative at the colony court and a lieutenant in the militia. On 20 June 1654 he was appointed lieutenant under Standish in the proposed expedition against the Dutch at Manhattan. On 2 October 1658 he was on the Council of war, served in 1671 as one of its chairmen. on 17 December 1673 he served as Surgeon General of the colony troops and the Massachusetts troops and was give 4 shillings a day.
During King Phillip's War he was captain of the Plymouth forces. Captain Fuller was also the first regular physician in Barnstable,. He lived at the northwest corner of Barnstable at Scorton Neck. He also owned large tracts of land in Falmouth and Middleboro.
His will was dated 25 July 1678 and was proved 30 October 1678. His estate was appraised at £1667,04,06, among the items were pearls, precious stones, and diamonds.
