Expression construct
The transgene expression cassette is flanked by AAV inverted terminal repeats (ITRs) with inclusion of the various regulatory elements. The components in green are essential and include the promoter, transgene and polyadenylation (poly A) marking the end of the transcriptional cassette. To improve transgene selectivity and efficiency, one can include cis-regulatory elements such enhancers, cis-regulatory modules (CRM), introns or post-transcriptional elements such as the woodchuck hepatitis virus post-transcriptional regulatory element (WPRE).
